More than 2 years teaching and demonstration experience of 3rd years and First year medical students, at Gunma University, Graduate School of Medicine.

Grants

Principal Investigator: Cysteine biosynthetic pathway is present in E. histolytica is a rational to develop chemotherapeutic agents against amebiasis is funded by Ministry of Education, Culture, Sports, Science and Technology (MEXT; www.mext.go.jp/english/) for two years from April 2007-March 2009. Co-investigator: Analysis of sulphur-containing amino acid metabolism and its regulation in E. histolytica is funded by MEXT from April 2006-2010.

Technical Skills

Cell Culture (Culture of Parasites, various bacterial and eukaryotic cell line).
Isolation, purification, and characterisation of proteins, glycoproteins and glycolipids.
Chromatography (gel-filtration, hydrophobic, thin layer, affinity and HPLC, FPLC).
Electrophoresis (PAGE, SDS-PAGE, Agarose, isoelectrofocusing).
Immunisation of animal for antibody production and identified efficacy of purified antigens in susceptible animal models (Hamster, mice, rat, and rabbit).
Preparation of various types of Liposomes and entrapment of antigen and drugs.
Immunological Techniques (B-cell, T-cell, Macrophage, Lymphocyte isolation and culture. ELISA and lymphocyte transformation test-LTT, Macrophage and promastigotes interaction).
Fluorescence, UV spectroscopy and confocal Microscopy.
In vitro and in vivo screening of drugs or compounds against pathogens. Many standard drugs or purified molecules tested both in vitro and in vivo using various liposome, adjuvant etc.
DNA: Extraction and purification of plasmid DNA, genomic DNA, total RNA, and mRNA. Enzyme digestion of DNA, PCR, RT-PCR, Colony-PCR, ARMS PCR, DNA sequencing, Ligation, Transformation, genomic DNA and cDNA cloning, protein expression in prokaryotes and eukaryotes, complementation, and over expression of proteins in eukaryotes, screening of anti-amoebic compounds against E. histolytica or transfected cell line. To develop in vitro drug resistance of parasites or cell lines.
Protein: Expression and purification in native (soluble) and /or denaturing condition by various methods, protein folding, protein structure and function determination, structural based drugs and/or inhibitor designing.
Southern, Northern & Western Hybridisation.
Enzyme assays, enzyme kinetics, enzyme inhibition (competitive, non-competitive, feedback and substrate), and evaluation of biochemical properties for various enzymes like stability, Temp., pH, cofactors, metal ion, substrate specificity, and develop new/modified enzyme assay system.
To evaluate protein-protein interaction and well familiar with using AKTA explorer (Pharmacia) to purified all macromolecules. Cellular fractional of phagosomes by sucrose and percoll gradient, retromer complex by chromatography for proteomic analysis.
Computer software program well known: MS Word, PowerPoint, Excel, Adobe Photoshop, Adobe Illustrator, Corel Graphic Suite (Coral Draw), Canvas, DNASIS, Genedoc, Endnote, phylogenetic analysis by Neighbor joining (NJ), Maximum Parsimony (MP), and MEGA3 etc.
